Optimizing of conditions for saponin extract process from Carica papaya L. leaves

Abstract

Papaya leaves are used to support for treatment of cancer in some countries such as Australia, American, Brasin, Vietnam… Papaya leaf contains alkaloid, saponin, carotenoid, phenolic compounds… In this study we optimise saponin extracted conditions from papaya leaves by using Design Expert program, and qualitative analyse saponin in papaya leaves. The total extract are extracted by MeOH. Three main factors are examined. There are extracted time, temperature and the ratio of material/extracted solvent. The research result show that optimised extracted conditions for total extract from papaya leaves are extracted time: 52.5 hours, the rate of material/solvent: 1/7 and extracted temperature: 48.51oC. Continue to extract saponin from the total extract with n-butanol, then purify by chloroform, we obtained total crude saponin with the content is 1,69 ± 0,37% compairing with dried material. Qualitative analyse saponins with foaming reaction and thin layer chromatography, we determine that saponins presented in papaya leaves is saponin steroid.
